Title: Advanced Flow Spectroscopy of Cells for Development of New Fluorophores

Abstract:

Fluorescent proteins (FPs) are powerful tools that permit real-time visualization of cellular processes. The utility of a particular FP for a specific experiment depends strongly on its brightness and photostability. However, the photon output of FPs is limited by dark-state conversion and irreversible photobleaching. Furthermore, FPs with emission red-shifted beyond 650 nm, would improve their utility for multicolor and whole-body imaging. These photophysical properties are directly related to the dynamics of the FP chromophore and the surrounding β-barrel. I will present a new microfluidicsbased selection strategy involving cell-based libraries of FPs that aim to modify the dynamics near the chromophore to develop new FPs with increased photostability. I will also present femtosecond spectroscopy and molecular dynamics investigations of hydrogen-bonding dynamics between the chromophore and its environment that provide insight into the mechanism of red-shifted emission. Variations on these new tools will lead to new opportunities for creating interesting and useful new biomolecules with selections and/or directed evolution based on molecular spectroscopy and dynamics.
